In the movie, it was stated that she is a citizen of Roran. Her husband was kidnapped and forced into labor, like many women and children in Roran, by Mukade, a rogue shinobi of Sunagakure (Hidden Sand Village). Unfortunately, this is the extent of the information available about her past and heritage.
In the movie, in an effort to get Sara (the last queen of Roran before it was destroyed) to free all the men who Mukade (the main antagonist) had forced into labor, she and the others abducted Sara. She told Naruto Uzumaki and Sara about their issue as he came in to help Sara.
Later, she is reunited with her husband when Sara frees the men who were laboring their lives away. Later, Masako and her husband follow Sarai, who first assists Sara in opening the gate to safety.
This is the only instance Masako ever appeared in the Naruto series. Based on this minimal appearance, Masako is believed to be alive and well until any additional information suggests otherwise.
