Ise and his wife Fuso was supplying relief to the suffering people during the Second Shinobi World War. Ise and his family were hiding in their destroyed home one night during the war when a Konoha ninja broke in seeking food.

They sought to elude capture but they were caught and to give Nagato a chance to flee, Ise battled the ninja. But the Konoha ninja, believing the pair to be rival ninjas, attacked and swiftly murdered both Fuso and the helpless Ise.

As a result of seeing this and accidentally reawakening his Rinnegan, Nagato murdered the two Konoha shinobi. Nagato laid his folks to rest before heading off to find nourishment. His parents' passing is the first major sorrow in his life and, in his opinion, proof that the peace Konoha advocates are achieved at the expense of other people's lives.

Ise was a kind person and a doctor who was eager to help everyone he could during times of conflict. He also had a deep affection for his son as he gave his life to let Nagato escape.
