Makino is first seen in the story working at a Party Bar serving Shanks, Luffy, and his friends. She and the mayor himself were the only two villagers who were brave enough to confront the bandit Higuma after he took Luffy after she went to search for the mayor. She seems to have brought some new clothes for Sabo and Ace while they were little and under Curly Dadan's care.
She appears to be being observed by numerous men, indicating that she is popular in the neighborhood, as she told Luffy to keep his plans from Garp a secret. Ace later asked her how to introduce Shanks to her after hearing so much about him from Luffy while carrying some imported beer to the bandit's cave. Makino was there when Luffy sailed away from their village on a little boat. She expressed her pain at leaving him.
After beating some of East Blue's most dreadful pirates, including Buggy, Don Krieg, and Arlong, Luffy was given his first bounty of 30,000,000 berries. Makino was working at an inn in Foosha Village at the time. At first, the locals celebrated the fame of one of their own, but Woop Slap stopped them, calling it dishonorable.
When Luffy received his bounty of 300,000,000 berries, Makino was once more seen talking with the mayor. She said that Luffy had made some wonderful friends and seemed to be having a great time.
Makino stopped Dadan from hitting Garp over Ace's death when Garp built a Marine Headquarters at Foosha Village to defend the village from the sudden increase in pirate and bandit activity following the Battle of Marineford. She said that Garp was grieving Ace's passing more than everyone else. Luffy was the one who was suffering the most, Dadan replied. This brings up a scene where she criticizes Ace for being impolite to Luffy and Ace. She started crying over this.
Makino gave birth to a baby while the Straw Hats were gone. She was spotted at Foosha Village with her infant in her arms as the locals celebrated Luffy's return.
With her infant in her arms and her eyes fixed on the ocean, Makino wondered how Luffy was faring. As soon as Luffy brought her the news about the happenings in Totto Land, Woop Slap surprised her by complaining that he had reappeared in the media.
Makino read some sad news about Sabo after the Levely. She shut down her bar for the day because she was so unhappy.
Makino agreed with Woop Slap's promise that Garp would be okay when residents of Foosha village responded to the news of Garp's missing on Hachinosu. She saw that her infant was starting to recognize Luffy's face from his frequent media appearances.
Makino is a very kind person. Makino seems to hold an important position in Foosha Village. Her love for Luffy gives her the appearance of being Luffy's adopted mother or sister, taking an interest in and encouraging whatever Luffy's ambition may be. She appears pleased with whatever kind of life Luffy leads as long as he is content and surrounded by friends.
Makino has black hair that reaches just over her shoulders and brown eyes. She is seen wearing a yellow kerchief, an orange shirt with black sleeves, a long light blue skirt, and orange trainers with white soles. When she met Ace for the first time she was seen wearing a pink-striped shirt underneath a white blouse with no sleeves. When she was young, she was seen wearing a bandana around her head that was yellow and covered more of her forehead.
Having known Luffy since he was a kid, Makino is familiar with him. Makino expressed her sadness at Luffy's departure and how much she would miss him. She has been following along with his shenanigans. When Dadan informed Makino that he was suffering the most following Ace's passing, she became very sad. After the time skip, Makino was happy to learn that Luffy had returned and that his bounty had increased.
Shanks and his crew are friendly with Makino; they've even visited her pub once.
When Ace and Luffy were kids, Makino treated Dadan as one of the regular customers at the Partys bar and frequently visited her home, bringing food and foreign beer for Dadan's group. After the Marineford War, she prevented Dadan from hurting Garp any further by claiming that Garp was also heartbroken by Ace's death.
Ace looked to Makino for advice on how to properly thank Shanks. Though he was rather irritated by her comment, Makino found it amusing. She felt sorrow over Ace's death as well.
Makino gets along well with Garp; while on Dadan's vacation, she frequently observes him disciplining Luffy and Ace. After the Marineford War, Makino defended Garp by saying that Garp also grieved Ace's death.
Makino is not only an anime character but is also a video game character as well.
